It all depends on where you want to get on the trail and what day you want to get there. The Metro-North train schedules are to be found here. The Hudson Valley line is great for anywhere from north of Bear Mountain to NY403. If you’re going to start at the Hudson, then go to Peekskill and take a taxi up to the bridge (best place to do that as its busier and has easiest access, and it’s a short cab ride and thus cheap.) If you’re starting on a weekend you can get an early train out (but check the schedule carefully as to which train on the line, since they don’t all go there) to Manitou, which only operates on weekends. From there it’s a very short walk to the trail (about .8 miles) along Manitou Rd. to the trailhead. If you’re headed further north, Garrison is good, though it’s a 2 mile road walk to the trail along NY403 (but it should be an easy hitch, if you don’t mind using your thumb.)
There are also other options depending on what you intend. If you’re headed SOBO, you might want to take a Short Line bus out of NYC instead, since they run service to Bear Mountain Inn. Short Line’s site is here, with daily service from the Port Authority to Bear Mountain. That would put you directly on the trail on the west side of the Hudson.
Hope all this helps, I try to encourage anyone who’s hiking the trail in the north-east to use as much public or mass transit as they can.

Strategic, excellent. Looks like I’ll take the train up to Peekskill on Friday and catch the train back to NYC from Pawling on Sunday on the Harlem line. That’s a nice hike!

Sounds like a good hike. I’ve had excellent experiences with Metro-North, very comfortable and efficient. And of course, catching a train at the Appalachian Trail Train Station is an experience not to be missed. You’ll have plenty of time for 46 miles between Friday morning and Sunday afternoon trains, but be careful of the schedule for your return, it’s limited at the AT stop so if you’re late on Sunday you’ll have to go on in to Pawling. Have fun with it all and good hiking!
